GLP-1对非酒精性脂肪肝大鼠TNF-α、TGF-β1及氧化应激的影响

摘要
目的探讨胰高血糖素样肽-1(GLP-1)对非酒精性脂肪肝(non-alconolic fatty liver disease,NAFLD)大鼠肿瘤坏死因子-α(TNF-α)、转换生长因子-β1(TGF-β1)及氧化应激的影响。方法雄性SD大鼠30只,随机分为普通喂养组(ND组)、高脂饮食组(HFD组)、高脂饮食长效人胰升糖素样肽(GLP-1)干预组(GLP-1组),喂养至16周末时处死全部大鼠,测定血清丙氨酸氨基转移酶(ALT)、TGF-β1、TNF-α、血清及肝组织匀浆甘油三酯(TG)、总胆固醇(TC),肝组织匀浆超氧化物歧化酶(SOD)、丙二醛(MAD)。结果体质量、肝指数及血清ALT、TG、TC、TGF-β1,HFD组明显高于ND组和GLP-1组(P<0.05),ND组与GLP-1组比较,差异无统计学意义(P>0.05);血清及肝组织匀浆中TG、TC、SOD、MDA、TNF-αHFD组均高于ND组和GLP-1组(P<0.05),GLP-1组与ND组比较均稍有升高,但差异无统计学意义(P>0.05)。结论 GLP-1能减轻NAFLD的脂肪变性,降低TNF-α及TGF-β1含量,改善氧化应激及脂质过氧化,提示使用GLP-1类似物利拉鲁肽等药物可作为治疗NAFLD的有效药物。
