普拉克索引起大鼠Y迷宫冲动样行为及机制

摘要
目的观察普拉克索(PPX)改变大鼠Y迷宫电击逃避任务的行为特点,并探讨其可能的作用机制。方法大鼠一次性sc给予PPX 0.1,1和10 mg·kg-1后,采用Noldus Etho Vision XT8影像行为轨迹分析系统检测大鼠Y迷宫电击逃避任务中判断正确反应次数、穿梭次数、运动距离和安全区停留时间的变化。通过大鼠震惊反射前脉冲抑制实验和清醒大鼠脑微透析实验,观察PPX对正常大鼠感觉运动门控系统以及对纹状体、杏仁核内单胺类神经递质含量的影响。结果与正常对照组对比,PPX组大鼠在Y迷宫判断正确反应次数上无显著性差异,但在一次逃避任务完成后、下一个任务开始前,迷宫内继续穿梭次数和运动距离显著增加(P<0.01),安全区停留时间明显减少(P<0.05),表现出类似强迫冲动的行为。震惊反射实验中,PPX组大鼠出现了前脉冲抑制受损的现象(P<0.01)。脑微透析结果显示,PPX对纹状体和杏仁核内多巴胺和5-羟色胺(5-HT)的浓度无明显影响。结论 PPX可能通过损害感觉门控系统而诱导大鼠表现出类似强迫-冲动的行为。
