共面多氯联苯ELISA方法的建立

摘要：

采用PCB半抗原Ⅰ与牛血清白蛋白的偶联物作为免疫抗原免疫新西兰大耳白兔,制备多克隆抗体并建立共面PCB的ELISA方法。结果表明,抗体的效价>6.4×105,建立的间接竞争ELISA法对PCB77、PCB126和PCB169检测的IC50值分别为18、31和186ng.mL-1,直接竞争ELISA法灵敏度高于间接竞争法,在ELISA反应液中添加4%DMSO,灵敏度最高。抗体对几种结构类似的共面PCB特异性从大到小排列顺序是:非邻位氯取代PCB、单邻位氯取代PCB和双邻位氯取代PCB。研究结果为PCB的快速检测提供了技术基础。
