基于UPLC-Q-TOF-MS技术的圣愈汤干预骨髓抑制大鼠的体内显效成分分析

摘要
目的:分析和表征圣愈汤干预骨髓抑制模型大鼠的入血成分,为其药效物质基础研究及质量控制提供依据。方法:利用大鼠体质量、血常规临床指标评价圣愈汤给药的有效性。以骨髓抑制SD大鼠为试验对象,灌胃给予圣愈汤,在显效状态下收集血浆样品,利用超高效液相色谱-四极杆-飞行时间串联质谱(UPLC-Q-TOF-MS)技术结合UNIFI系统分析表征吸收入血的原型成分及其代谢产物。结果:圣愈汤能够显著增加骨髓抑制大鼠的体质量、血红蛋白(HB)、红细胞计数(RBC)、血小板计数(PLT);在显效状态下,共表征出圣愈汤入血原型成分26个,代谢产物成分20个。主要血中移行成分包括梓醇、川芎内酯A、阿魏酸、芒柄花素、毛蕊异黄酮、人参皂苷Rb1、人参皂苷Rg1、人参皂苷Rg2、人参皂苷Rg3以及黄芪甲苷的代谢产物,具有对骨髓抑制的干预作用,为关键的活性成分。结论:研究结果初步阐释了圣愈汤干预骨髓抑制的药效物质基础,为圣愈汤药效物质研究和质量控制标准的提升提供科学依据。
