一氧化氮损伤神经细胞线粒体并诱导细胞凋亡

摘要
研究了一氧化氮对原代培养的大鼠小脑颗粒神经元的细胞毒性及其作用机理 .实验结果表明 ,一氧化氮供体S 亚硝基 N 乙酰基青霉胺 (SNAP ;5 0 0 μmol/L)可诱导未成熟小脑颗粒神经元凋亡 .流式细胞分析及HPLC分析表明 ,神经细胞经SNAP处理后线粒体跨膜电位及细胞内ATP含量均明显下降 .一氧化氮清除剂血红蛋白可有效保护线粒体 ,防止细胞凋亡 .实验结果提示 ,一氧化氮通过抑制线粒体呼吸链的活力、降低细胞内ATP含量而启动细胞凋亡程序 .
