人工改造的Cry1Ac和Cry1Ie基因在烟草中共表达对棉铃虫有更好的杀虫活性

摘要
利用叶盘转化法获得了携带Cry1Ac,Cry1Ie和同时携带这两个基因的3种类型转基因烟草.利用ELISA法测定转基因烟草叶片中Bt蛋白含量,在携带两种Bt基因的转基因烟草中,Cry1Ac和Cry1Ie蛋白分别占总蛋白的0.173%和0.131%.在携带单个基因的烟草中,Cry1Ac和Cry1Ie蛋白的含量分别为0.182%和0.124%.分别用野生型棉铃虫(Helicoverpa armigera)和Cry1Ac抗性棉铃虫的初孵幼虫在这3种类型的转基因烟草叶片上进行虫试,并以未转基因烟草为负对照.实验结果表明,携带Cry1Ie基因的转基因烟草对这两种棉铃虫都有杀虫活性,而携带Cry1Ac基因的转基因烟草仅对野生型棉铃虫有抗性,携带两种Bt基因的烟草比仅含其中一个基因的烟草对这两种棉铃虫均有更好的杀虫效果.该结果表明,这两种Bt基因在转基因烟草中能够协同作用,提高了烟草对害虫的杀虫效果.同时,转入两个Bt杀虫基因有可能成为延缓农业害虫对转基因作物产生抗性的新途径.
