7-硝基吲唑对缺氧缺血新生大鼠海马CA1区神经元凋亡的影响

摘要
目的 探讨选择性一氧化氮合酶 (NOS)抑制剂 7 硝基吲唑 (7 NI)对缺氧缺血新生大鼠海马CA1区神经元凋亡的影响。方法  7日龄Wistar大鼠 72只 ,随机分成 3组 ,(1)假手术组 :12只 ,仅作颈正中切口 ,不作颈总动脉结扎 ;(2 )观察组 (7 NI组 ) :30只 ,缺氧缺血性脑损伤 (HIBD)模型制作后即刻腹腔内注射 7 NI 2 5mg/kg ;(3)对照组 (生理盐水组 ) :30只 ,HIBD后即刻腹腔注射等体积的生理盐水。假手术组于手术后即刻 ,观察组、对照组于处置后 4 8小时断头取脑 ,分别进行HE和TUNEL染色 ,光镜下检测脑细胞凋亡数。结果 HE染色对照组大鼠海马CA1区可见典型的凋亡细胞 ,表现为细胞固缩 ,胞浆深染 ,核浓缩、碎片 ,核浆分布不清 ,有凋亡小体形成 ;7 NI组上述凋亡细胞明显减少 ;TUNEL染色对照组阳性细胞数与 7 NI组比较差异有极显著性 (P <0 .0 1)。结论  7 NI对HIBD后海马CA1区神经细胞凋亡有明显的抑制作用
