TFPI-2基因与消化道肿瘤关系的研究进展

摘要
组织因子途径抑制物2(tissue factor path-way inhibitor2,TFPI-2)是一种广谱的丝氨酸蛋白酶抑制物,可抑制包括纤溶酶、胰蛋白酶、基质金属蛋白酶(matrix metalloproteinases,MMPs)在内的多种蛋白酶,而MMPs参与了细胞外基质的重塑,在肿瘤细胞的浸润和转移、伤口愈合、血管新生、纤维蛋白溶解及动脉粥样硬化等生理和病理过程中发挥重要作用。TFPI-2可通过维持细胞外基质的完整、调节肿瘤血管生成、调控肿瘤细胞细胞凋亡及血液凝固等机制调节胰腺癌、食管癌及胃癌等消化道肿瘤细胞的侵袭转移。越来越多的研究表明,TFPI-2能明显抑制肿瘤中MMPs的活性与功能等从而抑制肿瘤的侵袭与转移,并成为肿瘤基因治疗的新靶点。
