磁性聚乙二醇载体固定化α淀粉酶的研究

摘要
A magnetic immobilized a-amylase (MIE) has been prepared by adsorbing α-amylase onto a magnetic collidal particle and cross-linking it with glutaraldehyde. The effects of different concentrations of glutaraldehyde, pH of buffers for immobilization and the properties of magnetic immobilization α-amylase were studied. The results showed that the activity of MIE was 34000 U per gram dry gel. The protein binding capacity was 100 mg per gram dry gel. The activity recovery was 37.7%. The optimum temperature of MIE was 30 degree higher than that of natural α-amylas(50℃). The optimum pH values of MIE was 1 pH unit higher than that of natUral α-amylase. The magnetic immobilized α-amylase has excellent thermal stability and working stability.
