OPG/RANK/RANKL信号通路在骨质疏松症中的研究进展和应用

骨保护蛋白(OPG)/核因子-Kβ受体活化因子(RANK)/核因子-Kβ受体活化因子配体(RANKL)信号通路是调节骨重建过程中破骨细胞功能的重要通路。成骨细胞释放的RANKL与破骨细胞表面的RANK结合后通过NF-Kβ途径、JNK途径、蛋白激酶B途径等促进破骨细胞的分化和激活;OPG则可竞争性抑制RANK与RANKL的结合,抑制破骨细胞功能,减少骨质破坏。OPG与RANKL的比例是调节骨吸收和骨形成平衡的重要因素。目前,该通路相关药物治疗骨质疏松的研究取得了一定的进展。本文对OPG/RANK/RANKL信号通路在骨循环中的作用机制和相关影响因素进行了综述,旨在为骨质疏松的防治提供新的思路。
