氢溴酸加兰他敏在小鼠血浆、脑组织中的药物动力学研究

摘要
建立了HPLC-荧光法测定小鼠血浆和脑组织中氢溴酸加兰他敏的浓度。采用IntersilC18柱,流动相为甲醇-水(40∶60,含0.2%三乙胺,磷酸调至pH6.4),激发波长和发射波长分别为290和320nm。小鼠单剂量口服氢溴酸加兰他敏4mg/kg后血浆和脑组织中的药物动力学参数tmax均为0.5h,Cmax分别为(691±80.34)和(1402±230.46)μg/L,AUC0→12h分别为(1138.2±160.5)和(2787.7±212.4)μg·h·L-1,AUC0→∞分别为(1147.01±198.94)和(2829.06±171.99)μg·h·L-1,t1/2分别为(4.6±1.4)和(5.0±0.2)h。
