骨髓基质细胞体外诱导分化为内皮细胞的超微结构研究

摘要
目的采用重组人粒细胞巨噬细胞集落刺激因子(rhGM-CSF)体外诱导骨髓基质细胞(BMSC)的方法,探讨BMSC向内皮细胞分化的可行性。方法在无菌条件下采集SD大鼠(雄性,体质量150~160g,10只)的长骨骨髓,获得骨髓细胞。实验组细胞以高质量浓度rhGM-CSF(400μg/L)为诱导剂,进行体外诱导分化;对照组培养基为常规1640培养液。培养至第8天,收集细胞后分别应用扫描电镜和透射电镜观察实验组分化后细胞的超微结构,并对分化后的细胞进行内皮细胞的鉴定。结果实验组细胞在透射电镜下可见内皮细胞结构特征及特征性的W-P小体结构,扫描电镜下可见细胞呈多边形,细胞膜表面有大量的细长丝状伪足及短小的微绒毛,符合内皮细胞的特点。结论BMSC在高浓度的rhGM-CSF诱导作用下分化的细胞具有内皮细胞的形态学特征。
