大鼠胆管阻塞性肝纤维化模型的建立

摘要
目的 建立大鼠胆管阻塞性肝纤维化模型 ,为肝纤维化机制和治疗研究提供基础。方法 雌性Wistar大鼠进行胆管内逆行性注入N 丁基 2 氰基丙烯酸盐加胆管结扎导致胆管阻塞 ,建立胆管阻塞性肝纤维化模型。观察手术后第 2 8d和 5 6d不同时间相关指标的改变。结果 胆管阻塞性肝纤维化模型ALT、AST、HA、LN均明显升高 ;病理组织学可见弥漫增生的胆小管 ,大量的胶原沉积在胆小管周围 ,肝细胞与增生的胆小管相比明显减少。结论 胆管阻塞性肝纤维化模型造模周期短 ,自发逆转率低 ,肝纤维化形成稳定 ,是研究肝纤维化的理想模型
