机械牵张作用对UMR-106细胞骨桥蛋白mRNA和TGF-β1 mRNA表达的影响

摘要
目的 探讨机械牵张作用对骨桥蛋白 (osteopontin ,OPN)mRNA和转化生长因子 β1(transforminggrowthfactorbeta 1,TGFβ1)mRNA表达的影响 ,说明正畸性骨改建的分子生物学机制。方法 建立体外培养细胞牵张施力模型 ,利用地高辛标记探针进行原位杂交 ,结合图象分析技术定量化研究基因表达的相对强度。结果 不同牵张强度和频率作用对OPNmRNA和TGF β1mRNA表达影响的差别具有显著性。低强度 低频率作用的影响最为显著 ,2 4h时OPNmRNA的表达强度从 0 16 13降低到 0 12 6 1(P <0 0 5 ) ;48h时TGF β1mRNA的表达强度从 0 0 899增加到 0 175 6 (P <0 0 1)。OPNmRNA和TGF β1mRNA的表达变化趋势相反。 结论 机械牵张作用可显著影响成骨细胞OPNmRNA和TGF β1mRNA的表达水平。低强度 低频率刺激对TGF β1mRNA表达的影响最显著 ,支持正畸临床所证明和提倡的持续轻力移动牙齿的原则。
