肿瘤放射治疗中辐射增敏剂的应用进展

摘要
肿瘤放射治疗中辐射增敏剂的作用机制主要包括改变肿瘤微环境、清除自由基和电子、细胞周期同步化、抑制DNA损伤修复、促进细胞凋亡和生物还原作用。目前临床应用的常规辐射增敏剂有硝基咪唑类化合物、环氧化酶-2抑制剂、铂类、天热药物,为减轻不良反应,采用小剂量多次照射或与解毒药物并用或局部用药等。新型辐射增敏剂有纳米粒、核仁素的特异性核酸适配体AS1411、STAT3反义寡核苷酸,他们作为放疗增敏剂能在肿瘤组织内被动靶向地聚集,进而达到杀死肿瘤的目的。近年来,寻找更高效辐射增敏剂的重点放在能影响某些还原酶的药物,设法进一步加重肿瘤乏氧状态,并且使提升细胞放射抗性的蛋白表达量下降,从而发挥细胞毒作用。
