槲皮素诱导TRAMP-C2细胞凋亡的实验研究

摘要
目的:探讨槲皮素对转基因前列腺腺癌小鼠C2细胞系(TRAMP-C2)凋亡影响及其作用机制。方法:采用不同浓度的槲皮素处理体外培养的TRAMP-C2细胞,应用MTT法检测槲皮素对TRAMP-C2细胞增殖的影响;采用流式细胞仪检测TRAMP-C2细胞的凋亡率,并应用基因芯片技术筛选槲皮素作用前后TRAMP-C2细胞差异性表达基因。结果:槲皮素能抑制TRAMP-C2细胞生长,诱导TRAMP-C2细胞凋亡,呈剂量及时间依赖性;基因芯片分析发现,应用5×10-5mol/L浓度槲皮素处理48h后的TRAMP-C2细胞有69个基因表达下调,94个基因表达上调,其中血小板反应素表达明显上调,为对照组的36倍。结论:槲皮素能诱导TRAMP-C2细胞凋亡,具有直接抗肿瘤作用,其作用机制可能与上调TRAMP-C2细胞血小板反应素-1的表达有关。
