细胞因子对神经干细胞增殖和分化的影响

摘要
目的 建立人胎大脑神经干细胞 (NSCs)分离培养的体外培养系统 ,鉴定其干细胞原始特征 ,观察上皮细胞生长因子(EGF)、碱性纤维母细胞生长因子 (FGF2 )和脑源神经生长因子 (BDNF)对NSCs分裂和分化的影响。方法 在含EGF和 (或 )FGF2的培养体系中分离培养NSCs。用定量RT PCR法检测干细胞巢蛋白表达丰度 ,用PCR ELISA法检测端粒酶活性 ,PC NA用免疫细胞化学染色检测干细胞增殖特征以及不同代数分裂的干细胞的比率。免疫细胞化学法证实NSCs分化后神经元和星形胶质细胞的比例趋势 (NF 2 0 0、MAP 2、synaptophysin、NeuN和GFAP染色 )。 结果 NSCs大量增殖呈球体状 ,PCNA阳性率高达 (71 6± 4 9) %、干细胞巢蛋白阳性、端粒酶活性为 6 3%。不同诱导因子对NSCs球体内细胞增殖数差异有显著性(P <0 0 5 )。证实EGF和 (或 )FGF2在培养基质存在下均能诱导NSCs分化 ,脑源神经生长因子 (BDNF)能维持神经元的存活和生长。分化后的神经元比例可达 (18 6± 2 5 ) %。结论 EGF和 (或 )FGF2可诱导NSCs分裂增殖并保持原始特性 ,在培养基质存在条件下 ,这些细胞因子可诱导NSCs分化。
