急性胰腺炎患者外周血单个核细胞p38 MAPK信号通路的变化

摘要
目的:探讨急性胰腺炎(acute pancreatitis,AP)患者外周血单个核细胞(peripheral blood mononuclear cell,PBMC)丝裂原活化蛋白激酶p38(p38 MAPK)信号水平的变化及其与疾病严重程度的关系。方法:收集轻型胰腺炎(mild acute pancreatitis,MAP)组29例及重症胰腺炎(severe acute pancreatitis,SAP)组23例,健康对照组21例。荧光实时定量PCR检测患者PBMC p38 MAPK基因表达水平,蛋白质印迹法检测PBMC p38 MAPK和磷酸化p38 MAPK(P-p38 MAPK)的水平。统计处理用单因素方差分析(One-Way Anova)和LSD法。结果:p38 MAPK基因表达水平,SAP组较对照组升高了11.7%(P<0.05),但MAP组与对照组比较差异无统计学意义;p38 MAPK总蛋白量SAP组较对照组增加了18.7%(P<0.05),但MAP组与对照组相比较差异无统计学意义;磷酸化p38 MAPK水平SAP组较对照组和MAP组分别提高了444.4%(P<0.01)和44.1%(P<0.05),MAP组较对照组增加了27.8%(P<0.05)。结论:p38 MAPK磷酸化水平在AP患者PBMC中显著升高,并与AP严重程度呈正相关;AP患者PBMC p38 MAPK基因表达水平和蛋白水平变化较小。
