多倍体山羊草物种中高分子量麦谷蛋白亚基的组成与其编码基因的分子克隆

摘要
山羊草属(Aegilops)是小麦属(Triticum)的近缘属之一, 山羊草属物种的种子中含有与普通小麦高分子量麦谷蛋白亚基(HMW glutenin subunit)结构相似的贮藏蛋白, 通称为山羊草高分子量麦谷蛋白亚基. 利用SDS-PAGE分析了4种多倍体山羊草物种所含高分子量麦谷蛋白亚基的组成, 之后又通过比较各种PCR扩增方法, 以偏凸山羊草(Ae. ventricosa)为材料建立了从多倍体山羊草物种基因组中扩增高分子量麦谷蛋白亚基编码基因的有效方法. 目前已从偏凸山羊草中克隆了两个高分子量麦谷蛋白亚基的编码基因, 氨基酸序列分析表明这两个基因分别编码1个x型和1个y型亚基.
