矽尘致肺纤维化中NO、NOS、TGF-β1、TNF-α的变化

摘要
目的 :探讨染矽尘大鼠体内NO含量、NOS活力、TGF - β1 和TNF -α含量的改变与矽肺的关系。方法 :采用一次性气管注射染矽尘方法建立动物模型 ;称量法测定脏器系数 ;硝酸还原酶法测定血浆、肺均浆NO含量 ;NOS催化L—Arg法测定血浆、肺匀浆NOS活力 ;采用ELISA法测定血浆、肺均浆TGF - β1 和TNF-α蛋白浓度 ;SAS6 1 2统计分析软件进行实验数据分析。结果 :染矽尘后第 7天、第 30天时 ,实验组肺匀浆NO (LNO)含量有显著升高 (P <0 0 1 ,P <0 0 5) ;实验组肺匀浆NOS (LNOS)活力在染矽尘后一直高于对照组 (P <0 0 1 )。血浆NO (PNO)含量仅在实验第 30天升高 ,而血浆NOS (PNOS)活力却在实验第 2 1天后低于对照组。实验组肺匀浆TGF - β1 (LTGF - β1 )在染矽尘后第 7天显著低于对照组 (P <0 0 1 ) ,第 1 4天显著高于对照组 (P <0 0 5) ;肺匀浆TNF -α (LTNF -α)在染矽尘后第 1 4天有显著性升高 (P <0 0 5) ,但在实验第 2 1天后 ,显著降低 (P <0 0 1 ) ;血浆TGF - β1 (PTGF - β1 )和TNF -α (PTNF -α)在实验过程中没有显著性差别。多元线性回归分析结果显示是否染尘、染尘时间、血浆TNF -α跟是肺纤维化有正相关关系 ,可能为肺纤维化的促进因素。而肺匀浆NOS、血浆TGF - β1 跟肺纤维化有负相关关系
