中药川芎对成骨细胞作用最佳浓度的体外实验

摘要
背景:已有研究报道中药川芎具有促进成骨细胞增殖及分化的作用。目的:进一步观察中药川芎中川芎嗪成分对成骨细胞增殖及Ⅰ型胶原蛋白表达的作用。方法:以小鼠成骨细胞株MC3T3-E1为研究对象,在1,5,10,15,20 mg/L 5个不同质量浓度川芎嗪干预下分别应用CCK-8方法测定成骨细胞增殖能力,酶联免疫法测定碱性磷酸酶活性及Ⅰ型胶原蛋白表达。结果与结论:(1)1,5,10,15,20 mg/L川芎嗪对成骨细胞的增殖均有促进作用,与空白对照组比较差异有显著性意义(P<0.005);(2)1,5,10,15,20 mg/L川芎嗪可以提高成骨细胞内碱性磷酸酶活性,其中1,5,10 mg/L川芎嗪组与空白对照组比较差异有显著性意义(P<0.005),15,20 mg/L川芎嗪组与空白对照组比较差异无显著性意义(P>0.005);(3)1,5,10,15,20 mg/L川芎嗪均促进Ⅰ型胶原蛋白表达,与空白对照组比较差异有显著性意义(P<0.005);(4)结果表明,中药川芎提取物川芎嗪对成骨细胞增殖及Ⅰ型胶原蛋白表达均有促进作用,质量浓度为10 mg/L效果理想。
