桃SSR-PCR主要因子对扩增产物的影响

以寿星桃为试材,通过Mg2+和DNA聚合酶、模板DNA和引物用量的两个双因素试验,以及dNTP用量的单因素试验,研究了5个主要因子对SSR扩增结果的影响,并对桃SSR反应体系进行了优化。结果表明,Mg2+和DNA聚合酶的用量对SSR扩增结果起着关键作用,且两者之间存在明显的互作效应;dNTP用量也对SSR的扩增结果起着重要作用,当浓度在200~300μmol/L时效果较好;模板DNA与引物用量之间没有很明显的互作效应,适当增加引物用量可以提高SSR的正确扩增;优化的桃SSR反应体系为:25μl的总反应体积中含1×PCR Buffe、Mg2+3 mmol/L、DNA聚合酶1.5 U、SSR引物各0.4μmol/L、模板DNA 50 ng、dNTP 200μmol/L。
