地黄醇提物及其药渣水提物抗抑郁作用的比较研究

摘要
目的比较地黄乙醇提取物(醇提物,EE)及其药渣水提物(RWE)的抗抑郁作用。方法雄性昆明小鼠随机分为对照组、阳性药盐酸氟西汀(FH,10 mg·kg-1)、地黄乙醇提取物及地黄药渣水提物的低、高剂量(2,4 g·kg-1,以生药计),连续灌胃(ig)给药7 d(每天1次),末次给药后1 h,进行开场实验(OFT)、悬尾实验(TST)、强迫游泳实验(FST)和利血平拮抗实验(RRT)测试;采用HPLC测定地黄提取物中主要药效物质梓醇含量。结果对照组小鼠在悬尾实验、强迫游泳实验的不动时间分别为(91.0±28.3)、(98.0±30.4)s,对照组在利血平拮抗实验的体温为(35.0±0.7)℃,地黄醇提物低剂量、高剂量(2,4g·kg-1)、药渣水提物高剂量(4 g·kg-1)和盐酸氟西汀(10 mg·kg-1)均显著缩短了不动时间(P<0.05)、逆转了体温降低(P<0.05)。开场实验中,模型组小鼠3 min内穿格次数为(91.9±24.6)次,各给药组与之相比均无显著差异;地黄乙醇提取物中梓醇含量为33.48 mg·g-1原药材,地黄药渣水提物中为0.034 mg·g-1原药材。结论地黄醇提物及其药渣水提物均有抗抑郁作用,其作用机制可能涉及单胺能神经系统。
