转谷氨酰胺酶2对乳腺癌耐药性的影响

摘要
目的探讨人乳腺癌耐药机制和转谷氨酰胺酶2(TG2)表达水平对化疗效果的影响。方法以TGM2慢病毒干扰载体(TGM2-RNAi-LV)构建转染细胞,设立3个组:RNAi组,转染沉默TGM2重组慢病毒颗粒;NC组,转染阴性对照病毒颗粒;Con组,仅在培养基中培养。Western blot检测MCF-7/ADR细胞TG2、Bcl-2和E-cadherin蛋白表达。3组MCF-7/ADR细胞加阿霉素处理,相应细胞分组为RNAi+Dox组、NC+Dox组和Dox组。TUNEL法检测阿霉素诱导肿瘤细胞凋亡,MTT法检测阿霉素对乳腺癌细胞作用的半数抑制浓度(IC50)。结果 RNAi组与NC组和Con组比较,TG2和Bcl-2表达水平明显下降,E-cadherin表达升高,差异有统计学意义(P<0.05)。RNAi+Dox组细胞的凋亡率为(31.37±1.30)%,与NC+Dox组和Dox组等比较差异有统计学意义(P<0.05)。RNAi+Dox组的IC50为(20.41±0.87)μmol/L,而NC+Dox组和Dox组的IC50分别为(40.68±1.23)μmol/L和(41.97±1.29)μmol/L,RNAi+Dox组的IC50明显低于NC+Dox组和Dox组(P<0.05)。结论RNA干扰TGM2表达,增强人乳腺癌细胞对阿霉素化疗的敏感性,下调TG2表达水平可能是预防乳腺癌患者耐药的一种新的方法。
