摘要：

根据已发表的PRRSV基因组序列 ,设计并合成了 1对特异扩增ATCCVR 2 3 3 2株的ORF6基因的引物。以ATCCVR 2 3 3 2基因组为模板 ,通过RT PCR扩增出 5 86bp的cDNA产物。将该cDNA片段经T A连接插入 pGEM Teasy载体中 ,用重组质粒转化大肠埃希氏菌JM 10 9,获得重组质粒转化菌。对重组质粒DNA进行PCR及酶切鉴定 ,证明插入的DNA片段与PCR扩增的DNA片段大小相符。对整个ORF6进行序列分析 ,将测序结果与已发表的ATCCVR 2 3 3 2序列相比较 ,证实插入片段为PRRSVVR 2 3 3 2的ORF6片段的cDNA。
