百里香醌通过抑制TLR4信号通路介导的炎性反应抗肝纤维化作用研究(英文)

摘要
目的:本实验通过体内、外实验研究百里香醌的抗肝纤维化作用及潜在的作用机制。方法:体内实验中,使用硫代乙酰胺(200mg/kg)建立小鼠肝纤维化模型,腹腔注射硫代乙酰胺每周3次,连续5周。百里香醌(20、40mg/kg)与硫代乙酰胺同时给药直至第5周,通过苏木素和伊红染色考察百里香醌的肝脏保护作用,然后提取肝脏组织蛋白以及mRNA用做蛋白印迹以及反转录-聚合酶链反应分析。体外实验中,肝星状细胞用不同浓度(3.125、6.25、12.5μg/mL)的百里香醌预处理1h后,加入1μg/mL内毒素24h后,提取细胞蛋白进行蛋白印迹分析。结果:体内实验结果显示,百里香醌能够有效对抗肝纤维化,并减少了由硫代乙酰胺刺激后造成的炎性细胞的浸润。百里香醌能够明显地降低Collagen-I,α-SMA以及TIMP-1的水平以。体外实验表明,百里香醌能够有效减少T-HSC/Cl-6的细胞活性。此外,百里香醌也能够明显降低由脂多糖(LPS)刺激的Collagen-I以及α-SMA的表达,抑制了TLR4、MyD88的水平以及NF-κB的核转导活性。结论:通过体内外实验证实百里香醌具有抗肝纤维化作用。这种潜在的作用机制与抑制Collagen-I和α-SMA的表达以及下调肝纤维化相关的炎性反应有关。此外,百里香醌能够抑制内毒素刺激的肝星状细胞中TLR4/MyD88的信号通路的激活。百里香醌在抗肝纤维化中具有潜在的治疗价值。
