贯叶连翘提取物对百草枯所致的急性肺微血管内皮细胞损伤的保护作用

摘要
目的探讨贯叶连翘提取物(HP)在百草枯(PQ)诱导的大鼠肺微血管内皮细胞(rPMVECs)急性损伤中的作用和对活性氧族物质(ROS)产生的影响。方法原代培养rPMVECs,分为PQ中毒组、HP干预组以及空白对照组。同时向PQ中毒组和HP干预组加入PQ溶液(0.1mmol/L)0.5h后开始计时,再向HP干预组分别添加三种不同浓度的HP溶液(250mg/L、10mg/L、1mg/L)。采用四氮唑盐(MTT)比色法观察不同时间点(3、12、24、48、72h)和不同浓度的HP对rPMVECs生长抑制率的影响,同时用分光光度比色法测定相应的细胞上清液中总超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果在0.1mmol/L的PQ诱导rPMVECs损伤后,随着HP浓度的增高和作用时间的增长,HP可明显减缓rPMVECs生长抑制率的升高;三种浓度的HP几乎在各个时间点均能明显减缓总SOD的活力的下降(P<0.05);72h时,三种浓度的HP均能明显减缓MDA含量的增加(P<0.05),而在其它时间点,三种浓度的HP几乎不能明显减缓MDA含量的增加。结论HP对PQ诱导的急性rPMVECs损伤具有保护作用。
