高迁移率族蛋白1联合白细胞介素-1β及脂多糖对类风湿关节炎患者成纤维样滑膜细胞增殖及炎症因子分泌的影响

摘要
目的探讨高迁移率族蛋白1(HMGBl)协同IL-1β及脂多糖对RA患者成纤维样滑膜细胞(FLS)增殖及炎症因子分泌的影响。方法收集在我院行关节置换或关节镜手术的RA患者的滑膜进行FLS的原代培养, 传代至4~6代用于实验。分别使用不同浓度HMGB1( 100、500、2 000 ng/ml), IL-1β (0.5 ng/ml)、脂多糖(100 ng/ml)单独刺激FLS;HMGBl/IL-1β混合物及HMGB1/脂多糖混合物共同刺激FLS。用细胞计数试剂盒(CCK-8)法检测FLS的增殖情况, ELISA法检测细胞上清中IL-6、IL-8、MMP-3的浓度。多样本间均数比较采用单因素方差分析。结果 HMGB1、IL-1β、脂多糖单独刺激和HMGBl/IL-1β、HMGB1/脂多糖混合物共同刺激都不影响FLS增殖(F=0.415,P=0.915)。除大剂量HMGB1组IL-6明显增高[(23.0±1.1) ng/ml]; HMGB1、IL-1β、脂多糖单独刺激不影响FLS分泌IL-6、IL-8和MMP-3,但HMGB1与IL-1β、脂多糖共同刺激可明显促进FLS分泌IL-6、IL-8和MMP-3 (F值分别是97.804, 117.383, 70.179, P=0.000),且呈HMGB1剂量依赖性。结论 HMGB1与IL-1β、脂多糖共同刺激有明显的促炎因子活性, 可显著增加IL-6、IL-8、MMP-3的产生。
