达格列净对心肌肥厚和心肌纤维化的影响研究

摘要
目的探讨达格列净对腹主动脉缩窄(AC)压力超负荷大鼠模型的心肌肥厚和心肌纤维化的影响。方法将60只雄性SD大鼠分为假手术(Sham)组、Sham+达格列净组、AC组、AC+达格列净组,通过建立腹AC压力超负荷模型,12周后检测心脏功能、心肌肥厚和心肌纤维化情况,以及心肌组织氧化应激、炎性反应相应指标。结果与AC组比较,AC+达格列净组左心室舒张末期压力(LVEDP)降低[(9.03±2.65)mm Hg vs.(16.93±3.11)mm Hg],等容舒张期左心室压力下降最大变化率(-dP/dt)升高[(7 886±547)mm Hg/s vs.(6 257±467)mm Hg/s],左心室重量指数(LVMI)降低[(3.520±0.108)vs.(4.351±0.116)],心肌细胞面积缩小[(335.7±27.6)μm2 vs.(398.5±35.6)μm2],胶原容积分数(CVF)降低[(7.54±0.89)%vs.(13.65±1.62)%],差异有统计学意义(P<0.05)。同时,与AC组比较,AC+达格列净组丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β、IL-6、单核细胞趋化蛋白-1(MCP-1)水平降低,超氧化物歧化酶(SOD)水平升高,差异有统计学意义(P<0.05)。结论达格列净可减轻心肌氧化应激和炎性反应来抑制心肌肥厚及心肌纤维化。
