血塞通注射液对局灶性脑梗死大鼠NgR mRNA及蛋白表达的影响

摘要
目的:观察血塞通注射液在不同时点对局灶性脑梗死大鼠Nogo-A受体(NgR)蛋白及mRNA表达的影响,揭示血塞通注射液对脑缺血受损神经元的重塑作用机制。方法:采用改良Zea-Longa方法制备MCAO模型,SD大鼠35只随机等分为假手术组、模型组、血塞通小剂量组、血塞通大剂量组、尼莫地平组。各组动物分别于术后7天、14天、28天麻醉断头取材,行免疫组织化学及荧光定量PCR检测。结果:模型组NgR蛋白及mRNA表达随着缺血时间延长而逐渐升高,到28天时达到高峰,较同时间点假手术组比较均增高(P<0.05);各用药组均比模型组低,术后14天及28天,血塞通大剂量组较模型组显著降低(P<0.05)。结论:NgR参与了脑梗死后突触的再生抑制,血塞通注射液可以抑制NgR蛋白及mRNA表达,从而促进神经再生。
