急性脑梗死患者外周血单核细胞TLR4表达及其与TNF-α、IL-6的相关性

目的观察急性脑梗死患者外周血单核细胞Toll样受体4(TLR4)的表达及其与TNF-α、IL-6浓度的相关性,初步探讨TLR4在急性脑梗死缺血性炎性损伤中的作用,为临床治疗脑梗死提供新的思路。方法分别运用Tapman实时定量PCR及流式细胞仪法检测39例急性脑梗死患者外周血单核细胞(淋巴细胞、单核细胞)TLR4 mRNA、蛋白的表达,用ELISA法测定血清TNF-α、IL-6的浓度,与35例同期正常对照及32例脑动脉粥样硬化患者比较。结果脑梗死组发病第3天TLR4表达、血清TNF-α、IL-6浓度均显著高于正常对照组和动脉粥样硬化组(P<0.01)。TLR4表达与血清炎性因子TNF-α、IL-6浓度变化呈正相关(P<0.01)。结论研究提示TLR4可能在急性脑梗死缺血性炎症性损伤中具有重要的作用,其机制可能是通过上调TLR4表达促使炎性因子产生分泌来介导脑缺血炎性损伤。
