表皮细胞生长因子对小鼠非酒精性脂肪肝炎TNF-α、CHOP的影响

摘要
目的探讨表皮生长因子(EGF)对小鼠细胞非酒精性脂肪性肝炎(NASH)模型炎症因子及内质网应激的影响。方法用蛋氨酸与胆碱缺乏(MCD)的培养基诱导AML12小鼠正常肝细胞株建立NASH模型,分别设空白对照组、NASH造模组、EGF组,Western blot检测各组肿瘤坏死因子α(TNF-α)、C/EBP同源蛋白(CHOP)蛋白表达,油红O染色检测细胞内脂质沉积程度,实时定量荧光PCR(real-time PCR)检测TNF-α、CHOP的mRNA转录水平。结果 EGF组TNF-α、CHOP的蛋白及mRNA相对表达量与NASH组比较显著下降(P<0.05),脂质沉积变化不明显。结论 EGF能明显下调NASH中TNF-α、CHOP的表达水平,但对脂质沉积无明显改变。
