人参皂苷Rh2抗小鼠哮喘模型气道炎性反应的作用研究

摘要
目的:探讨人参皂苷Rh2对小鼠哮喘模型气道炎性反应影响的作用机制。方法:雌性BALB/c小鼠50只随机分成5组,即正常对照组,哮喘模型组,人参皂苷Rh2低、高剂量组、地塞米松组。在末次激发24h后所有小鼠取左肺组织行苏木精-伊红(HE)染色。分别采用酶联免疫吸附法(ELISA)和蛋白质印迹检测支气管肺泡灌洗液(BALF)中白细胞介素(IL)-4、IL-5、IL-13含量以及肺组织中磷酸化AKT、p38MAPK及核转录因子-κB(NF-κB)p65蛋白的变化。结果:与正常对照组比较,小鼠哮喘模型组BALF中炎性细胞计数、IL-4、IL-5、IL-13水平升高(P<0.01);肺组织中磷酸化AKT、p38 MAPK及NF-κB p65蛋白表达水平升高(P<0.05)。与哮喘模型组比较,人参皂苷Rh2高剂量组小鼠BALF中炎性细胞计数、IL-4、IL-5、IL-13水平和肺组织中磷酸化AKT、p38MAPK及NF-κB p65蛋白表达水平均明显降低(P<0.05)。人参皂苷Rh2处理能明显减轻哮喘小鼠肺组织病理学改变,人参皂苷Rh2低、高剂量干预组之间比较,无显著差异。结论:人参皂苷Rh2对小鼠哮喘具有保护作用,其机制可能部分与抑制磷酸化的AKT、p38MAPK以及NF-κB信号通路有关。
