槲皮素改善TNF-α诱导的肠上皮Caco-2细胞功能障碍

摘要
为研究槲皮素改善TNF-α诱导的肠屏障功能障碍的细胞模型作用,实验采用TNF-α200 ng/mL孵育Caco-2单层细胞24 h构建上皮屏障功能障碍的模型,再采用槲皮素(5,15,30μmol/L)干预模型,考察槲皮素对上皮单层细胞通透性及上皮的紧密连接(TJ)蛋白Claudin-1和Occludin蛋白功能及表达的影响。结果表明,与正常对照组比,TNF-α组的上皮通透性增强,屏障功能障碍,且TJ蛋白中的Claudin-1、Occludin的磷酸化成分减少。槲皮素干预模型细胞后浓度依赖地增强上皮屏障功能,上调Claudin-1、Occludin的磷酸化成分及Occludin的整体蛋白成分。提示槲皮素可通过加强紧密连接蛋白的结构和功能改善肠上皮屏障功能。
