C反应蛋白基因单核苷酸多态性和环境交互作用与缺血性脑卒中的相关性分析

摘要
目的探讨C反应蛋白(CRP)基因单核苷酸多态性(SNP)(rs1130864和rs1800947)和环境交互作用与缺血性脑卒中(IS)发生的相关性。方法采用病例对照研究,连续收集IS病人236例和非IS对照者270例,采用PCR-RFLP对CRP基因SNP(rs1130864和rs1800947)进行基因分型。应用χ2检验分析CRP基因SNP与IS及其亚型的相关性。采用unPHASED软件进行单体型分析。以多因素Logistic回归分析SNP与环境因素间交互作用对IS发生的影响。结果 IS组与对照组rs1130864基因型、等位基因频率分布比较差异无统计学意义。IS组rs1800947-GC基因型频率高于对照组[OR=2.205,95%CI(1.251,3.887),P=0.005]。两个SNP构建单体型分析显示,IS组与对照组单体型C-C频率分布存在统计学意义[OR=2.392,95%CI(1.325,4.320),P=0.007]。Logistic回归分析显示,吸烟人群携带rs1800947-GC基因型IS发生风险升高[OR=6.276,95%CI(2.288,17.221),P<0.001],饮酒人群携带rs1800947-GC基因型IS发生风险亦升高[OR=9.207,95%CI(2.489,34.056),P=0.001]。结论 CRP基因rs1800947-GC基因型、单体型C-C可作为人群IS的遗传标志物,rs1800947与吸烟、饮酒存在显著的交互作用;而rs1130864与IS的发生风险无关。
