加味牛蒡子汤促进TNF-α转基因小鼠淋巴管回流功能防治类风湿关节炎的实验研究

摘要
目的观察加味牛蒡子汤(JWNBZT)对类风湿关节炎(RA)模型动物TNF-α转基因(TNF-Tg)小鼠关节炎症和关节周围淋巴回流功能的影响。方法将16只清洁级3月龄左右TNF-Tg小鼠按照体重随机分为TNF-Tg组和JWNBZT组,每组8只。另取10只同窝对照体重相似的野生型小鼠(WT)作为对照组,JWNBZT组每天以JWNBZT灌胃,对照组和TNF-Tg组每天以等量纯净水灌胃,治疗12周后小鼠足底皮下注射吲哚菁绿(ICG)测量小鼠淋巴管回流功能,并取踝关节制作石蜡切片进行苏木素-伊红(HE)染色观察炎性浸润和关节骨质破坏情况,抗酒石酸酸性磷酸酶(TRAP)染色观察破骨细胞数量。结果与对照组比较,TNF-Tg组小鼠淋巴管回流功能出现明显损害,Clearance和Pulse明显下降,淋巴管渗漏面积增多,踝关节内出现明显炎症浸润且骨质减少,距骨内破骨细胞增多(P<0.05);与TNF-Tg组比较,JWNBZT组小鼠淋巴管回流功能明显增强,Clearance和Pulse升高,淋巴管渗漏面积减少,踝关节内炎症浸润减轻且骨质增多,距骨内破骨细胞减少(P<0.05)。结论 JWNBZT能够减轻RA受累关节的炎症浸润和关节骨质破坏,并促进淋巴管回流功能,这可能是其治疗RA的机制。
