丹参酮ⅡA对小鼠压力超负荷诱导心肌损伤保护作用研究

摘要
目的探讨丹参酮ⅡA(TanⅡA)对小鼠压力负荷性心肌损伤保护作用。方法选取30只雄性C57BL/6小鼠为研究对象,将所有小鼠随机分成对照组、模型组及TanⅡA组,每组各10只。采用升主动脉缩窄构建小鼠压力负荷心肌损伤模型,术后TanⅡA组给予TanⅡA,连续3周,第4周处死所有小鼠,收集标本。测量小鼠心脏重量和心脏重量指数,酶联免疫吸附法和蛋白印迹法检测炎症和凋亡相关蛋白表达。结果与对照组比较,模型组小鼠心脏重量和心脏重量指数显著增加,白介素(IL)-1β、肿瘤坏死因子-α(TNF-α)、IL-6、Bax、Caspase-3蛋白表达升高,IL-10、Bcl-2表达降低,差异有统计学意义(P<0.05);与模型组比较,TanⅡA组小鼠心脏重量和心脏重量指数显著降低,IL-1β、TNF-α、IL-6、Bax、Caspase-3蛋白表达降低,IL-10、Bcl-2表达升高,差异有统计学意义(P<0.05)。结论 TanⅡA处理对小鼠压力负荷性心肌损伤有保护作用,其作用机制可能通过调节炎症反应和凋亡实现。
