二烯丙基硫化物对百草枯中毒急性肺损伤大鼠的抗氧化作用

摘要
目的探讨二烯丙基硫化物(DAS)对百草枯(PQ)中毒大鼠急性肺损伤的影响。方法 100只雄性Wistar大鼠随机分为5组,正常对照组、PQ 70 mg·kg-1模型组、DAS 25,50和100 mg·kg-1治疗组,每组20只。一次性ig给予PQ 70 mg·kg-1制备PQ中毒模型,DAS组于造模前、后30 min ip给予相应剂量药物,正常对照组ip给予等量生理盐水。各组分别于造模后1,3,6和12 h麻醉大鼠,取右肺下叶进行HE染色,光镜下观察肺组织病理损伤程度并评分;右肺上叶进行肺组织一氧化氮(NO)含量检测;左肺进行支气管肺泡灌洗获取肺泡巨噬细胞(AM),培养24 h取上清,分光光度法测定诱导型一氧化氮合酶(i NOS)含量;培养至72 h,采用细胞免疫化学方法检测AM中i NOS蛋白的表达。结果与正常对照组比较,模型组肺泡结构破坏严重,病理评分明显升高(P<0.01),肺组织中NO含量明显升高(P<0.01),AM培养上清液中i NOS含量明显升高(P<0.01),i NOS蛋白表达增加(P<0.05);与模型组比较,DAS 50 mg·kg-1组在造模后3,6和12 h以及DAS 100 mg·kg-1组在造模后1,3,6和12 h肺泡结构损伤显著减轻,病理评分降低(P<0.05);DAS25和50 mg·kg-1组肺组织中NO和i NOS含量及i NOS蛋白表达减少(P<0.05),DAS 100 mg·kg-1组NO含量减少更为明显(P<0.01)。结论 DAS可能通过抑制PQ中毒大鼠肺组织中的氧化损伤因子减轻肺损伤。
