食品级二氧化钛对幼鼠肠道菌群及血清炎性因子的影响

摘要
目的研究食品级二氧化钛(titanium dioxide,TiO2)对幼鼠肠道菌群的影响及血清炎性因子的影响。方法将24只无特定病原体(specific pathogen free,SPF)雄性ICR小鼠随机分为三组,分别为对照(生理盐水)组和低(10 mg/kg TiO2)、高(40 mg/kg TiO2)剂量组,每组8只。采用灌胃方式进行染毒,每天一次,连续染毒14 d;收集幼鼠粪便,采用高通量测序技术对各组幼鼠肠道菌群进行分析; ELISA法测定C反应蛋白(CRP)、白介素1α(IL-1α)、白介素1β(IL-β)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)和单核细胞趋化蛋白-1(MCP-1)水平; HE染色观察结肠组织病理学变化。结果与对照组相比,高剂量组Chao1指数、OTU数量显著降低,拟杆菌门和Prevotella显著升高(P<0.05);与低剂量组相比,高剂量组Tenericutes门增高(P<0.05);高剂量组CRP、IL-1α、IL-1β、TNF-α和MCP-1水平高于对照组(P<0.05);各组结肠组织出现不同程度的病理学改变。结论食品级TiO2暴露可导致幼鼠肠道菌群变化以及出现轻度的炎性反应。
