摘要
为了更直接、更方便检测乳酸杆菌表达系统,研究通过PCR法以质粒pEGFP-C1为模板扩增出增强型绿色荧光蛋白基因(egfp)序列,将扩增产物连接到乳酸杆菌的整合型载体pMJ67上,成功构建了重组质粒pMJ-egfp。将质粒pMJ-egfp电转化入乳酸杆菌L.casei CECT5276,egfp编码序列通过质粒上的Lac操纵子同源重组整合到L.casei CECT5276染色体上,连续传代50次,发现绿色荧光蛋白能够稳定遗传。说明试验获得了能在无抗性选择压力下稳定表达绿色荧光蛋白的重组菌L.casei CECT5278。
