视黄酸相关孤儿受体-白介素-23信号通路诱导病理性Th17细胞分化介导血压升高机制研究

摘要
目的:探讨视黄酸相关孤儿受体(RORγt)-白介素-23(IL-23)信号通路在病理性辅助性T细胞17(Th17)分化及盐敏感性高血压(SSHT)发病中的免疫机制,揭示RORγt-IL-23R信号通路通过调节Th17细胞的功能在高血压中的作用。方法:收集临床病例共90例,根据盐负荷试验,筛选盐敏感性高血压患者、非盐敏感性高血压患者及正常血压人群健康志愿者。流式细胞技术检测CD4+Th17细胞占辅助性T细胞(CD4+T细胞)的比例。ELISA检测外周血中TH17分泌白介素-17A(IL-17A)比例。检测正常环境、不同浓度NaCl(10、20、40、80 mmol/L)下人初始CD4+T细胞中Th17细胞比例及外周血中IL-17A比例,40 mmol/L NaCl下CD4+T细胞加入RORγt拮抗剂、IL-23R拮抗剂后测Th17细胞比例。结果:盐敏感性高血压组CD4+Th17细胞占CD4+T细胞的比例明显高于健康志愿者组。不同浓度NaCl培养人初始CD4+T细胞,CD4+Th17细胞占CD4+T细胞的比例均高于健康志愿者组,并且随着NaCl浓度增高,CD4+Th17细胞占CD4+T细胞的比例逐渐增高。高浓度NaCl培养基下CD4+T细胞中分别加入RORγt拮抗剂、IL-23R拮抗剂,CD4+Th17细胞占CD4+T细胞的比例均较40 mmol/L NaCl培养人初始CD4+T细胞组CD4+Th17细胞占CD4+T细胞的比例降低。结论:TH17细胞引起的免疫反应参与了高血压的进程,促进SSHT的发生。RORγt-IL-23R信号通路可诱导病理性Th17细胞分化从而导致SSHT的发生。
