白癜风患者外周血中miR-155、miR-1238-3p、miR-202-3p、miR-630和miR-766-3p的表达与病情进展的研究

摘要
目的本研究旨在探讨miR-155、miR-1238-3p、miR-202-3p、miR-630和miR-766-3p在白癜风患者外周血中的表达改变与病程发展的关系。方法本次研究选取2018年1月—2019年1月来我院治疗和检查的62例白癜风患者,平均年龄(23.25±2.11)岁,男30例(48.39%),女32例(51.61%),其中白癜风稳定期患者44例(71.77%),白癜风进展期患者18例(28.23%)。选取与白癜风组性别、年龄和体质量指数匹配的健康者作为对照组;通过实时荧光定量PCR(RT-qPCR)和蛋白质免疫印迹(Western blot)检测受试者外周血中miR-155、miR-1238-3p、miR-202-3p、mi R-630和miR-766-3p的表达量;通过酶联免疫吸附测定(ELISA)法检测肿瘤坏死因子(TNF)-α、干扰素(IFN)-γ、IFN-α和白细胞介素(IL)-1β蛋白水平含量;通过活性氧检测试剂盒和流式细胞仪检测受试者白癜风患者细胞内活性氧含量。结果随着白癜风病程发展,miR-155 m RNA和蛋白表达量逐渐增加,mi R-1238-3p、miR-202-3p、mi R-630和miR-766-3p mRNA和蛋白表达量逐渐降低(P<0.05)。白癜风进展期miR-155 mRNA和蛋白表达量高于白癜风稳定期,mi R-1238-3p、miR-202-3p、miR-630和miR-766-3p mRNA和蛋白表达量低于白癜风稳定期(P<0.05);相较于健康对照组,白癜风稳定期和白癜风进展期均能使TNF-α、IFN-γ、IFN-α和IL-1β含量增加,白癜风进展期TNF-α、IFN-γ、IFN-α和IL-1β含量高于白癜风稳定期;相较于健康对照组(2.36±0.15),白癜风稳定期(3.85±0.47)和白癜风进展期(8.18±0.56)活性氧含量均增加,且进展期高于稳定期(P<0.05)。结论随着白癜风病程发展,外周血中mi R-155表达量逐渐增加,miR-1238-3p、miR-202-3p、miR-630和miR-766-3p表达量逐渐降低。
