放射后肝癌细胞MMP-2表达、活性的变化及其对侵袭性的影响

摘要
目的探讨放射后肝癌细胞侵袭潜能的变化及其机制。方法以不同剂量(0、4、8Gy)高能 X 线照射人肝癌低转移潜能细胞株 MHCC97L。明胶酶谱法检测放射后24、48、72、96 h 细胞培养上清液金属蛋白酶(MMP)-2活性,细胞免疫荧光法检测放射后24、48和72 h 细胞内 MMP-2蛋白表达,Transwell 法测定放射后48和96 h 细胞的侵袭能力。结果放射后细胞 MMP-2蛋白分泌及活性增强并与放射剂量及时间相关,8 Gy 放射后48 h MMP-2蛋白活性最高,达到对照组的2倍。0、4、8 Gy 放射后48 h 穿过 transwell 小室的细胞数分别为7.2±1.9、13.2±2.7和31.2±7.2(P<0.05),放射后96 h 组间差异无统计学意义。结论放射后肝癌细胞侵袭潜能短暂增强,放射诱导细胞 MMP-2分泌及活性增强是侵袭性增强的原因之一。
