洋水仙花粉和花粉原生质体中肌动蛋白微丝分布变化的共焦显微镜观察

摘要
用荧光标记的鬼笔环肽作为肌动蛋白微丝的探针,结合共焦显微镜技术,观察了洋水仙(Narcissus cyclamineus)花粉及花粉原生质体中微丝骨架的分布变化情形。在水合而未萌发的花粉粒中,存在于周质的微丝束多数与花粉粒的长轴呈直角排列。但在萌发沟部位,微丝束的排列则呈网状。在花粉粒的周质至中央部位,微丝束形成另一种形式的网络,在结构上比在萌发沟部位的网络疏松。当花粉开始萌发时,存在于花粉粒内的微丝束逐步转变为平行排列状,并向花粉管伸长的方向延伸和聚集。在脱壁后的花粉原生质体内,微丝束经过重组形成一个结构复杂和排列紧密的网络。原生质体经低渗爆破处理或Triton X-100等过程抽提离析,在遗留下来的膜层上仍然可以见到微丝束残迹,显示微丝与膜层关系密切。用双染法同时显示微管和微丝,表明两者的分布有重叠现象,说明微管和微丝骨架的分布并不是随机的,而可能是由同一种结构或机制控制着。
