带J链的α-防御素-1的基因重组和表达载体的构建

摘要
目的 将α防御素 1(HNP 1)改建为一种杀菌分子J链α防御素 1(J HNP 1) ,这种杀菌分子具有与细胞膜上具备的多聚IgA受体(PIgR)相结合而进入粘膜上皮细胞的能力,发挥其杀菌作用。即克隆人Jchain HNP 1嵌合基因DNA ,并构建真核表达载体。方法 将从pCH质粒中获取的J链片段,采用人工连接方法与HNP 1片断相连接,并将其构建于新型哺乳动物细胞质粒表达载体pcDNA3 .1( ) Myc HisC中,进行序列测定。结果 经过重组得到的J链防御素 1的目的片断为786bp ,序列分析与GenBank中报道的编码J链及HNP 1成熟肽的序列完全一致。结论 Jchain HNP 1嵌合基因DNA的克隆和哺乳动物细胞表达载体的构建,为进一步进行抗菌肽的研究及制备奠定了基础。
