茄子IRAP分子标记体系的建立与优化

摘要
为确保茄子IRAP反应结果的稳定性和重复性,进而为茄子遗传多样性分析和品种鉴定提供可靠的新方法,按照已报道的马铃薯和烟草的反转录转座子LTRs保守区域设计IRAP引物,以茄子品系HL-01为试材,对影响茄子IRAP反应的重要因素进行了初步研究,建立并优化了茄子IRAP分子标记技术体系。优化的茄子IRAP分子标记体系为:20(l反应液中,10×反应buffer(100mmol/L Tris-HCl,100mmol/L KCl,80mmol/L(NH4)2SO4,pH9.0,NP-40)2μl,2mmol/L MgCl2,0.2mmol/L dNTPs,Taq-l.0U,0.4μmol/L引物,DNA模板50ng。PCR反应程序为:94℃预变性4min,然后按94℃变性30s,45℃退火30s,72℃延伸2min,进行32个循环,最后72℃延伸10min。
