呋喃它酮代谢物人工抗原及多克隆抗体的制备

摘要
本文利用对醛基苯甲酸(CPA)与呋喃它酮代谢物(AMOZ)进行衍生化,得到衍生物CPAMOZ,并通过质谱法对其结构进行确证。然后采用混合酸酐法将CPAMOZ与载体蛋白偶联,采用紫外扫描(UV)法对偶联物进行验证。用偶联成功的人工全抗原(CPAMOZ-BSA)免疫BALB/c小鼠,间接ELISA测定小鼠血清抗体效价为1∶8×104,对NPAMOZ的半数抑制量为26.55μg/L,间接竞争ELISA鉴定小鼠血清多克隆抗体,与NPAMOZ、CPAMOZ的交叉反应率分别为100%、44.8%,与AMOZ、对醛基苯甲酸、对硝基苯甲醛以及同类其他三种药物呋喃妥因、呋喃西林、呋喃唑酮等原药及其代谢物以及代谢物衍生物的交叉反应均小于0.01%。结果表明,制备的AMOZ多克隆抗体效价高、特异性强,灵敏度高,为进一步制备AMOZ单克隆抗体和研制快速筛选检测试剂盒奠定了基础。
