巨峰葡萄查尔酮异构酶基因克隆及表达分析

摘要
从一年生‘巨峰’扦插苗中克隆了CHI基因,并采用半定量RT-PCR技术,以actin基因为内参,分析了CHI基因在‘巨峰’葡萄果实不同发育阶段及组织的表达。序列分析表明:CHI基因序列中含有3个内含子,4个外显子。信息学分析显示,CHI基因编码的多肽链既没有信号肽,也不存在跨膜结构域,且亚细胞定位于细胞质中。半定量RT-PCR分析结果表明,CHI基因在果皮、果肉、种子及幼叶和幼根中都表达,但不同的组织中表达方式不同。CHI基因在花后30 d和90 d的果皮中强烈表达;但在果肉中,CHI基因在花后90 d表达强烈,而在种子中于花后45 d表达强烈,随果实发育,其表达逐渐下降。在上述实验的同时,高温处理抑制CHI基因在一年生盆栽‘巨峰’葡萄幼叶中的表达,随处理时间延长,其表达又有所恢复;高温诱导CHI基因在幼根的表达。
