葛根素对缺氧复氧心脏微血管内皮细胞的保护作用

摘要
目的:探讨葛根素在心脏微血管内皮细胞(cardiac microvascular endothelial cell,CMEC)缺氧复氧损伤中的作用。方法:将CMEC随机分为空白组、模型组、葛根素低、中、高剂量(25,50,100 mg·L-1)共3组。四甲基偶氮唑(methylthiazolyl tetrazolium,MTT)法检测细胞活力,流式细胞仪检测细胞凋亡,比色法检测乳酸脱氢酶(lactate dehydrogenase,LDH)活性、丙二醛(malondialdehyde,MDA)含量及超氧化物歧化酶(superoxide dismutase,SOD)活性,Western blot法检测Caspase-3和Bcl-2蛋白表达。结果:葛根素预处理后,缺氧复氧CMEC细胞活力增强,细胞凋亡率、LDH活性及MDA含量降低,SOD活性增强,Caspase-3蛋白表达减少,Bcl-2蛋白表达增加。结论:葛根素可拮抗缺氧复氧所致的CMEC损伤,可能与其抗氧化及抗凋亡活性相关。
