液相色谱质谱联用技术及其在药物代谢研究中的应用

摘要
近 2 0多年来 ,液相色谱 质谱 (LC MS)联用技术已发展成为一种高度自动化、常规的应用分析手段 ,在各个领域得到了广泛应用。各种软离子化技术 ,特别是电喷雾(ESP)、离子喷雾 (ISP)、大气压化学电离 (APCI)的引入 ,使LC MS对高极性化合物的分析具独特优势 ,在药物代谢研究中发挥着日趋重要的作用 ,在很大程度上替代了传统的气相色谱 质谱 (GC MS)联用技术 ,简化了样品 (特别是结合型代谢物 )处理过程。本文就LC MS存在的问题、仪器接口、离子化方法、流动相组成、MS的发展及其在药物代谢物研究中的应用等方面作一简要总结 ,重点介绍了各种不同接口的构造和原理
